1. MDX-010

MDX-010 is designed to increase immunity to cancer. It is a laboratory-produced antibody to the cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) protein found on certain lymphocytes (a type of white blood cell). When lymphocytes recognize a foreign substance, such as a virus or bacteria, they initiate an immune response to fight and control the infection. Once this is achieved, CTLA-4 protein help stop the immune response, decreasing the number of immune cells against the virus or bacteria. When an immune response is mounted against tumor cells, however, it may be beneficial not to stop the immune response, but instead, to keep a large number of lymphocytes available to recognize and fight tumor cells. MDX-010 blocks CTLA-4 protein and thus increases the immune response.

2. MDX-060

MDX is a fully human antibody that targets CD30, a member of the tumor necrosis factor receptor superfamily and a molecule found on activated lymphocytes. CD30 has been found to be over-expressed in several lymphoproliferative disorders and is present on malignant cells of Hodgkin's lymphoma and anaplastic large cell lymphoma (ALCL), as well as other CD30-positive lymphomas.

3. MDX-070

MDX-070 is a fully human antibody that targets prostate specific membrane antigen (PSMA), a cell surface marker that is preferentially expressed on malignant prostate tissues. Preclinical studies using human prostatic cancer cells suggest that MDX-070 may effectively target prostate tumor cells in patients.

4. MDX-214

MDX-214 targets the epidermal growth factor receptor (EGFR), a receptor molecule implicated in tumor cell growth and found to be over-expressed in a variety of cancers, including head and neck, breast, colon, prostate, lung and ovarian cancers. MDX-214 provides an unusually potent approach to treating EGFR-positive cancers by inhibiting tumor cell growth and by mediating antibody-dependent cellular cytotoxicity (ADCC).
